Одним из широко используемых специфических препаратов моноклональных антител c anti-HER2 (human epidermal growth factor receptor 2) активностью является трастузумаб – это высокоэффективное средство для «удаления» гиперэкспрессированных единиц рецептора HER2 с поверхности клеток и снижения его онкогенности. Как и для других биологических лекарственных препаратов, для трастузумаба одной из возможных нежелательных реакций со стороны иммунной системы является иммуногенность – выработка противолекарственных антител к препарату.</p></sec><sec><title>Цель</title><p>Цель. Разработка и валидация методики полуколичественного определения антител к трастузумабу в сыворотке крови человека.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Полуколичественное определение антител проводилось методом иммуноферментного анализа в сочетании с техникой ACE, с использованием спектрофотометрического детектирования в видимом диапазоне спектра.</p></sec><sec><title>Результаты и обсуждения</title><p>Результаты и обсуждения. Разработанная методика была валидирована по показателям: предел исключения, селективность, чувствительность, «хук»-эффект, толерантность к присутствию лекарственного препарата, прецизионность и стабильность (краткосрочная и долгосрочная). Для снижения интерференции компонентов биологической матрицы в анализе на этапе разработки было определено значение минимального необходимого разбавления (1 : 10). Рассчитанные значения предела исключения для этапа скрининга (фактор нормализации) и подтверждающего анализа составили 0,004 и 34,59 %, соответственно. Чувствительность разработанной методики составила 99,5 нг/мл антител к трастузумабу.</p></sec><sec><title>Заключение</title><p>Заключение. Полученные при валидации методики результаты позволяют применять методику полуколичественного определения антител к трастузумабу в сыворотке крови человека для проведения аналитической части исследований безопасности препаратов трастузумаба.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Introduction</title><p>Introduction. One of the widely used specific anti-HER2 (human epidermal growth factor receptor 2) MAb drugs is trastuzumab. Trastuzumab is highly effective for malignant HER2 hyperexpression reduction, which results in HER2 oncogenicity decrease. As any other biotherapeutics trastuzumab can cause immunological adverse reactions, e.g. immunogenicity or anti-drug antibodies (ADAs) production.</p></sec><sec><title>Aim</title><p>Aim. The aim of this study was to develop and validate the analytical method for anti-trastuzumab antibodies determination in human blood serum.</p></sec><sec><title>Materials and methods</title><p>Materials and methods. The semi-quantitative anti-trastuzumab antibody determination was carried out by the ELISA method combined with ACE technique, using spectrophotometric detection in the visible range of the spectrum.</p></sec><sec><title>Results and discussion</title><p>Results and discussion. The developed method was validated for cut point, selectivity, sensitivity, "hook" effect, drug tolerance, precision and stability (short-term and long-term). To decrease the background noise from non-specific binding of sera components, the minimum required dilution value was determined at 10 % serum. The calculated values for screening cut point (normalization factor) and confirmatory cut point were 0.004 and 34.59 %, respectively. The sensitivity of the developed method was estimated at 99.5 ng/mL of anti-trastuzumab antibodies.</p></sec><sec><title>Conclusion</title><p>Conclusion.
