Эторикоксиб – селективный ингибитор циклооксигеназы (ЦОГ-2), применяемый для лечения острой боли и обладающий противовоспалительной и анальгетической эффективностью. Эторикоксиб вызывает меньшее количество осложнений по сравнению с другими нестероидными противовоспалительными препаратами (НПВП). ФБУ «ГИЛС и НП» была разработана глазная жидкая лекарственная форма на основе эторикоксиба. В настоящей статье предложена методика определения содержания эторикоксиба в жидкой лекарственной форме методом высокоэффективной жидкостной хроматографии с УФ-детектированием.</p></sec><sec><title>Цель</title><p>Цель. Разработка и валидация методики количественного определения эторикоксиба в жидкой лекарственной форме.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Для анализа использовали глазные капли с концентрацией действующего вещества эторикоксиба 0,05 %, стандартный образец эторикоксиб (Kekule Pharma Limited, Индия, серия ACE-3 WS001/15). Хроматографическое разделение проводили на высокоэффективном жидкостном хроматографе Agilent 1220 Infinity II LC (Agilent Technologies, США), оснащенном градиентным насосом, термостатом колонок и детектором с диодной матрицей. Анализ проводили на колонке Kromasil С8 250 × 4,6 мм, используя в качестве подвижной фазы ацетонитрил и 0,05 М буферный раствор дигидрофосфата калия рН = 4,2 в соотношении 46 : 54. Время анализа составило 15 минут при длине волны детектирования 235 нм.</p></sec><sec><title>Результаты и обсуждение</title><p>Результаты и обсуждение. Разработана и проведена валидация методики количественного определения эторикоксиба в жидкой лекарственной форме по следующим показателям специфичность, линейность, правильность, промежуточная (внутрилабораторная) прецизионность, повторяемость.</p></sec><sec><title>Заключение</title><p>Заключение. По результатам валидационных испытаний все перечисленные параметры соответствуют критериям приемлемости. Предлагаемая методика характеризуется высокой эффективностью и специфичностью.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Introduction</title><p>Introduction. Etoricoxib is a selective cyclooxygenase (COX-2) inhibitor used for the treatment of acute pain and has anti-inflammatory and analgesic efficacy. Etoricoxib causes fewer complications compared to other non-steroidal anti-inflammatory drugs (NSAIDs). FSI "SID and GP" has developed an ophthalmic liquid dosage form based on etoricoxib. This article proposes a method for determining the content of etoricoxib in a liquid dosage form by high performance liquid chromatography with UV detection.</p></sec><sec><title>Aim</title><p>Aim. Development and validation of a method for the quantitative determination of etoricoxib in liquid dosage form.</p></sec><sec><title>Materials and methods</title><p>Materials and methods. Eye drops with a concentration of the active substance etoricoxib of 0.05 % were used for the analysis, a standard sample of etoricoxib (Kekule Pharma Limited, India, series ACE-3 WS001/15). Chromatographic separation performed on an Agilent 1220 Infinity II LC high performance liquid chromatograph (Agilent Technologies, USA) equipped with a gradient pump, a column thermostat, and a diode array detector. The analysis carried out on a Kromasil C8 column 250 × 4.6 mm, using acetonitrile and 0.05 M buffer solution of potassium dihydrogen phosphate pH = 4.2 as a mobile phase in a ratio of 46 : 54. The analysis time was 15 minutes at a detection wavelength of 235 nm.</p></sec><sec><title>Results and discussion</title><p>Results and discussion. A method for the quantitative determination of etoricoxib in a liquid dosage form developed and validated according to the following indicators: specificity, linearity, accuracy, range, intermediate precision, repeatability.</p></sec><sec><title>Conclusion</title><p>Conclusion. According to the results of validation tests, all of the listed parameters meet the acceptance criteria.
