Радиорезистентность раковых клеток серьезная проблема при лучевой терапии опухолевых заболеваний. Радиосенсибилизирующие препараты делают злокачественные клетки более чувствительными к излучению и повышают эффективность лучевой терапии, однако их широкое клиническое применение ограничено существенными побочными эффектами. Разработка и изучение новых радиосенсибилизаторов представляется актуальной задачей современной фармакологии.</p></sec><sec><title>Цель</title><p>Цель. Целью данной работы было изучение эффективности аскорбата лития в качестве радиосенсибилизатора при воздействии фотонного и нейтронного излучения в широком диапазоне доз.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Оценка биологического действия выполнялась на опухолевой линии аденокарциномы рака предстательной железы PC-3. В качестве генераторов ионизирующего излучения использовался циклотрон для получения нейтронного потока и источник кобальт-60 для получения гамма-излучения.</p></sec><sec><title>Результаты и обсуждение</title><p>Результаты и обсуждение. Доказано усиление цитотоксического эффекта при сочетанном применении разных видов ионизирующих излучений и аскорбата лития. Выявлена устойчивость лини рака предстательной железы к гамма-излучению в поглощенной дозе 0,5–3,0 Гр. Показано, что опухолевые клетки рака предстательной железы более чувствительны к воздействию исследуемого препарата в минимальных концентрациях в сочетании с нейтронным облучением в сравнении с гамма-излучением в одинаковой поглощенной дозе. Основной механизм радиосенсибилизирующего действия аскорбата лития заключается в локальной индукции окислительного стресса, синергетически усиливающий действие ионизирующего излучения.</p></sec><sec><title>Заключение</title><p>Заключение. Сочетание аскорбата лития с нейтронным излучением приводит к более выраженному результирующему цитотоксическому эффекту. Повышение концентрации аскорбата лития приводило к нарастанию прооксидатного эффекта с усилением повреждающего действия на клетки.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Introduction</title><p>Introduction. Radioresistance of cancer cells is a serious problem in radiation therapy of tumor diseases. Radiosensitizers make malignant cells more sensitive to radiation and increase the effectiveness of radiation therapy; however, their widespread clinical use is limited by significant side effects. The development and study of new radiosensitizers seems to be an urgent task of modern pharmacology.</p></sec><sec><title>Aim</title><p>Aim. The purpose of this work was to study the effectiveness of lithium ascorbate as a radiosensitizer under the influence of photon and neutron radiation in wide dose range.</p></sec><sec><title>Materials and methods</title><p>Materials and methods. Evaluation of the biological effect was carried out using the tumor line of prostate cancer PC-3. We used a cyclotron to produce neutron radiation and a Cobalt-60 source to produce gamma radiation.</p></sec><sec><title>Results and discussion</title><p>Results and discussion. We have proved an increase in the cytotoxic effect with the combined use of different types of ionizing radiation and lithium ascorbate. The resistance of the prostate cancer line to gamma radiation at an absorbed dose of 0.5–3.0 Gy was revealed. It was shown that tumor cells of prostate cancer are more sensitive to the effects of the study drug in minimal concentrations in combination with neutron irradiation compared to gamma radiation in the same absorbed dose. The main mechanism of the radiosensitizing action of lithium ascorbate is the local induction of oxidative stress, which synergistically enhances the action of ionizing radiation.</p></sec><sec><title>Conclusion</title><p>Conclusion. The combination of lithium ascorbate with neutron radiation leads to a more pronounced resulting cytotoxic effect.
