Одним из эффективных подходов к получению инъекционных форм труднорастворимых лекарственных веществ является получение их наносуспензий, стабилизированных альбумином. Примером успешного применения этой технологии является препарат Abraxane® (nab™-паклитаксел, от англ. Nanoparticle albumin-bound, Celgene Corporation). В настоящем исследовании этот подход использован для получения инъекционной формы труднорастворимого пролекарства этопозида.</p></sec><sec><title>Цель</title><p>Цель. Получение инъекционной формы нового 4’-О-бензилоксикарбонильного производного этопозида (ETP-cbz) на основе человеческого сывороточного альбумина и изучение ее цитотоксичности на клетках рака молочной железы (РМЖ).</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Структуру ETP-cbz подтверждали методами ЯМР-спектроскопии и элементного анализа. Значения log P рассчитаны in silico с использованием Molinspiration Cheminformatics. Наночастицы ETP-cbz, стабилизированные альбумином (ETP-cbz-HSA), получены методом гомогенизации под давлением. Размер, распределение частиц по размерам и концентрацию дезинтеграции определяли методами динамического рассеивания света (ДРС) и атомно-силовой микроскопии (АСМ). Содержание ETP-cbz и альбумина в форме определяли методом ВЭЖХ. Конверсию ETP-cbz в этопозид изучали в модельной среде в присутствии эстеразы. Цитотоксичность ETP-cbz-HSA в сравнении с этопозидом и свободным ETP-cbz оценивали на клетках РМЖ (HBL-100 и MCF-7) при помощи колориметрического МТТ-теста.</p></sec><sec><title>Результаты и обсуждение</title><p>Результаты и обсуждение. Синтезировано новое производное этопозида, обладающее более высокой по сравнению с этопозидом гидрофобностью (log P 2,42 и 0,7 соответственно). Благодаря лабильной карбонатной связи, которая легко расщепляется под действием эстеразы, ETP-cbz является пролекарством этопозида. Разработан метод получения наноформы ETP-cbz на основе альбумина. Содержание ETP-cbz в суспензии составило 1,75 мг/мл, размер частиц – 110 ± 9 нм (ДРС), концентрация дезинтеграции – 3,0 ± 0,1 мкг/мл. Цитотоксичность наноформы ETP-cbz в отношении клеток MCF-7 и HBL-100 in vitro сопоставима с активностью свободного этопозида и ETP-cbz.</p></sec><sec><title>Заключение</title><p>Заключение. Полученная наноформа пролекарства этопозида на основе альбумина, пригодная для инъекционного введения, проявила высокую цитотоксичность в отношении клеток РМЖ in vitro и заслуживает дальнейшего изучения для оценки возможности ее применения в химиотерапии РМЖ.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Introduction</title><p>Introduction. One of the promising strategies for the development of injectable formulations of poorly water-soluble drugs is the preparation of their nanosuspensions stabilized with albumin. A well-known example of the successful implementation of this technology is Abraxane® (nanoparticle albumin-bound paclitaxel). In the present study, this approach is used for the development of the injectable formulation of a hydrophobic etoposide prodrug.</p></sec><sec><title>Aim</title><p>Aim. Development of the injectable formulation of a novel 4'-О-benzyloxycarbonyl derivative of etoposide (ETP-cbz) based on human serum albumin and evaluation of its cytotoxicity against breast carcinoma cells (BC).</p></sec><sec><title>Materials and methods</title><p>Materials and methods. The structure of ETP-cbz was confirmed by NMR spectroscopy and elemental analysis; log P was calculated in silico using Molinspiration Cheminformatics. The ETP-cbz-HSA nanoparticles were prepared by high pressure homogenization. The particle size, size distribution, and disintegration concentration were evaluated by dynamic light scattering (DLS) and atomic force microscopy (AFM). The contents of ETP-cbz and albumin in the formulation were determined by HPLC. The conversion ETP-cbz-HSA to etoposide was studied in a model medium in the presence of esterase. The cytotoxicity of ETP-cbz-HSA compared to etoposide and free ETP-cbz was assessed on BC cells (HBL-100 and MCF-7) using a colorimetric MTT assay.</p></sec><sec><title>Results and discussion</title><p>Results and discussion. ETP-cbz exhibited higher hydrophobicity as compared to the etoposide (log P 2.42 vs 0.7, respectively). Due to easy cleavage of the carbonate bond in the presence of esterase, ETP-cbz can be considered an etoposide prodrug. The ETP-cbz-containing HSA nanoparticles prepared by a high-pressure homogenization technique had the particle size of 110 ± 9 nm (DLS) and a low disintegration concentration of 3.0 ± 0.1 μg/mL. The drug content was 1.75 mg/mL. The nanoformulation demonstrated high cytotoxic activity in vitro against MCF-7 and HBL-100 cells that was comparable to the activity of substances etoposide and ETP-cbz.</p></sec><sec><title>Conclusion</title><p>Conclusion.
