Агонисты α2-адренорецепторов могут быть использованы не только как антигипертензивные и седативные средства, но также представляют большой интерес для лечения неврологических нарушений. В ранее проведенных исследованиях для представителя данной группы 6-оксо-1-фенил-2-(фениламино)-1,6-дигидропиримидин-4-ола (мафедина) была показана высокая эффективность в качестве нейропротекторного средства. Однако его влияние на поведение животных в поведенческих тестах еще не было оценено.</p></sec><sec><title>Цель</title><p>Цель. Целью настоящей работы стала оценка влияния натриевой соли мафедина в трех дозах (1, 10 и 50 мг/кг) на поведение белых беспородных мышей в 3 тестах: «Открытое поле», «Приподнятый крестообразный лабиринт» и «Черно-белая камера».</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Эксперименты были выполнены на 60 белых беспородных мышах-самцах массой 20–22 г, рандомизированных на 4 группы (n = 15): 1) контроль (0,9%-й раствор натрия хлорида); 2) мафедин (1 мг/кг); 3) мафедин (10 мг/кг); 4) мафедин (50 мг/кг). Все препараты вводили однократно внутрибрюшинно за 20 мин до тестирования. Поведение животных оценивали в тестах «Открытое поле», «Приподнятый крестообразный лабиринт» и «Черно-белая камера» по общепринятым методикам с перераспределением по группам после каждого теста и интервалом между тестами не менее 2 суток. Статистическую обработку данных проводили с использованием пакета программного обеспечения Prism 8.0.2.</p></sec><sec><title>Результаты и обсуждение</title><p>Результаты и обсуждение. Мафедин в дозах 1 и 10 мг/кг не оказывал влияние на поведение испытуемых животных ни в одном из тестов. Доза 50 мг/кг оказывала на испытуемых животных анксиолитическое действие, о чем свидетельствовали снижение индекса тревожности в тесте «Приподнятый крестообразный лабиринт» и увеличение количества выглядываний из черной камеры в тесте «Черно-белая камера» по сравнению с контрольной группой.</p></sec><sec><title>Заключение</title><p>Заключение. Натриевая соль мафедина в диапазоне доз от 1 до 50 мг/кг не оказывает негативного влияния на поведение мышей, что свидетельствует о высоком профиле безопасности изучаемого соединения. Обнаруженное противотревожное действие высокой дозы изучаемого соединения определяет интерес к его изучению не только как нейропротекторного, но и противотревожного средства.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Introduction</title><p>Introduction. α2-adrenergic agonists are not only used as antihypertensive and sedative agents, but are also of interest as potential medications for the treatment of neurological disorders. Previous research has shown a compound from this class, 6-oxo-1-phenyl-2-(phenylamino)-1,6-dihydropyrimidine-4-ol (mafedine), to exert strong neuroprotection under experimental conditions. Despite its long record of development, the effects of mafedine on animal behavioural characteristics remain unknown.</p></sec><sec><title>Aim</title><p>Aim. This work was aimed at evaluating the effects of mafedine sodium at three doses (1, 10, or 50 mg/kg) on white outbred mouse behavior in three tests: Open Field, Elevated Plus Maze, and Light/Dark Box.</p></sec><sec><title>Materials and methods</title><p>Materials and methods. Experiments were carried out on 60 white outbred male mice weighing 20–22 g, randomized into 4 groups (n = 15): 1) control (0,9 % saline); 2) mafedine (1 mg/kg); 3) mafedine (10 mg/kg); 4) mafedine (50 mg/kg). All agents were administered via single intraperitoneal injections 20 min before testing. Animal behavior was assessed using the Open Field, Elevated Plus Maze, and Light/Dark Box tests following conventional protocols with group reassignment between tests and an inter-test time interval of at least 2 days. Statistical analysis was carried out using the Prism 8.0.2 software package.</p></sec><sec><title>Results and discussion</title><p>Results and discussion. At 1 or 10 mg/kg, mafedine did not affect animal behaviour in either of the tests. At 50 mg/kg, it produced an anxiolytic effect, as indicated by the decrease in the anxiety index values for the Elevated Plus Maze test as well as the increase in peeking out frequency in the Light/Dark Box test, compared to respective control values.</p></sec><sec><title>Сonclusion</title><p>Сonclusion. Mafedine sodium salt at doses between 1 and 50 mg/kg was shown to produce no adverse effect on mouse behaviour, indicating a good safety profile of the compound.
