Новый селективный ингибитор PAR-2-рецепторов, 3-(2-бутил-5-хлоро-1Н-имидазол-4-ил)-N-[4-метокси-3-(трифторметил)фенил]-4,5-дигидро-1,2-оксазол-5-карбоксамид (R004), находится на стадии доклинического исследования. Изучение экскреции R004 и его метаболитов ранее не проводилось.</p></sec><sec><title>Цель</title><p>Цель. Изучение экскреции R004 и его метаболитов с мочой и калом после однократного перорального и внутрибрюшинного введения субстанции.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Исследование проводилось на 2 группах крыс линии Wistar по 6 особей. Первой группе субстанция R004 вводилась перорально в дозировке 10 мг/кг, второй группе – внутрибрюшинно в дозировке 10 мг/кг. Забор биоматериала проводился с помощью метаболических клеток. Пробы мочи отбирались до введения препарата и в промежутки 0–4, 4–8, 8–12, 12–24, 24–48, 48–72, 72–96, 96–120 ч после введения. Пробы фекалий отбирались до введения препарата и в промежутки 0–12, 12–24, 24–48, 48–72, 72–96, 96–120 ч. Анализ образцов проводился с помощью ВЭЖХ-МС/МС.</p></sec><sec><title>Результаты и обсуждение</title><p>Результаты и обсуждение. Аналитический диапазон методики определения R004 и 4-метокси-3-(трифторметил)-анилина (М2) в моче составил 5–2000 нг/мл, а 3-(2-бутил-5-хлоро-1Н-имидазол-4-ил)-4,5-дигидро-1,2-оксазол-5-карбоновой кислоты (М1) – 100–40 000 нг/мл. В фекалиях концентрации R004 измеряли в диапазоне 0,5–500,0 мкг/г, M1 – 4–4000 нг/г, М2 – 40–40 000 нг/г. Основная часть лекарственного средства и метаболитов выводится в течение 48 ч после введения. Полная элиминация достигается спустя 96 ч. R004 экскретируется в неизменном виде только с фекалиями: 94,83 ± 0,78 % при пероральном введении и 57,63 ± 0,66 % при внутрибюшинном введении (М + m). Метаболит М1 выводится преимущественно ренальным путём, метаболит М2 – преимущественно через кишечник.</p></sec><sec><title>Заключение</title><p>Заключение. Биоаналитические методики определения R004, М1 и M2 в моче и кале были успешно валидированы. Большая часть R004 экскретируется в неизменённом виде энтеральным путем. М1 выводится преимущественно с мочой, М2 – с фекалиями.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Introduction</title><p>Introduction. The new selective inhibitor of PAR-2 receptors, 3-(2-butyl-5-chloro-1H-imidazole-4-yl)-N-[4-methoxy-3-(trifluoromethyl)phenyl]-4,5-dihydro-1,2-oxazole-5-carboxamide (R004), is at the stage of preclinical trail. The excretion of R004 and its metabolites has not been studied before.</p></sec><sec><title>Aim</title><p>Aim. Investigation of excretion of R004 and its metabolites in urine and feces after a single oral and intraperitoneal administration of substance.</p></sec><sec><title>Materials and methods</title><p>Materials and methods. The study was carried out on 2 groups of 6 Wistar rats. The R004 substance was administered orally to the first group at a dosage of 10 mg/kg, to the second group intraperitoneally at a dosage of 10 mg/kg. Biomaterial sampling was carried out with using metabolic cages. Urine was collected before administration of the drug and in the intervals of 0–4, 4–8, 8–12, 12–24, 24–48, 48–72, 72–96, 96–120 h after administration. Feces were collected before administration of the drug and in the intervals of 0–12, 12–24, 24–48, 48–72, 72–96, 96–120 h. The samples were analyzed using HPLC-MS/MS.</p></sec><sec><title>Results and discussion</title><p>Results and discussion. The analytical range of the urine quantification method for R004 and 4-methoxy-3-(trifluoromethyl) aniline (М2) was 5–2000 ng/ml, and 3-(2-butyl-5-chloro-1H-imidazole-4-yl)-4,5-dihydro-1,2-oxazole-5-carboxylic acid (M1) – 100–40 000 ng/ml. In feces concentrations of R004 were measured in the range of 0.5–500.0 µg/g, M1 – 4–4000 ng/g, M2 – 40–40 000 ng/g. The main part of the drug and metabolites was excreted within 48 h after administration. Complete elimination was achieved after 96 h. R004 is excreted unchanged only with feces: 94.83 ± 0.78 % after oral administration and 67.04 ± 0.59 % after intraperitoneal administration (M + m). The metabolite M1 is mainly excreted by renal route, the metabolite M2 is mainly excreted through intestine.</p></sec><sec><title>Conclusion</title><p>Conclusion. Bioanalytical methods for determination of R004, M1 and M2 in urine and feces have been successfully validated. Most part of the R004 is eliminated unchanged by enteric route.
