На международном фармацевтическом рынке важное место занимают биопрепараты, созданные с использованием методов рекомбинантной ДНК. Сегодня бактериальные штаммы и клеточные линии млекопитающих широко используются для производства рекомбинантных белков, моноклональных антител и вакцин. Для того чтобы поддерживать постоянное качество и однородность исходного посевного материала, создаются банки клеток.</p></sec><sec><title>Текст</title><p>Текст. Основные рекомендации по разработке банков клеток изложены в руководствах, касающихся качества лекарственных препаратов, ICH Q5. В России аналогичные требования представлены в решении Совета Евразийской экономической комиссии № 89 «Об утверждении Правил проведения исследований биологических лекарственных средств Евразийского экономического союза». В соответствии с вышеуказанными рекомендациями возможно формирование одно- или двухуровневой системы банка клеток. Первым шагом должно быть создание главного банка клеток (ГБК), который создается в соответствии с высокими стандартами качества. ГБК следует инициировать из одной хорошо охарактеризованной бактериальной колонии. Следующим шагом будет создание рабочего банка клеток (РБК) из одного или нескольких контейнеров ГБК. В связи с тем, что все РБК созданы на основе хорошо охарактеризованных ГБК, их характеризация может быть сокращена. При характеризации оценивается идентичность, чистота и стабильность банков клеток. Рекомендации для проведения тестов представлены в нормативных документах. Тесты на идентичность подтверждают, что клетки в банке точно идентифицированы и соответствуют предполагаемой клеточной линии. Это крайне важно для предотвращения перекрестной контаминации и обеспечения использования соответствующих клеточных линий в производственных процессах. Оценка чистоты проводится с целью подтверждения отсутствия контаминантов, таких как бактерии, грибы и микоплазмы, в составе клеточной линии. Контроль микробиологической чистоты является ключевым компонентом оценки и гарантирует, что в производственном процессе используются клетки, свободные от контаминантов. Характеризация клеточных банков позволяет оценить генетическую стабильность клеток с течением времени. Это включает в себя оценку того, сохраняют ли клетки свои предполагаемые генетические характеристики и не подвергаются ли нежелательным мутациям во время культивирования. При трансфере технологии производитель также должен предоставить полную информацию о характеризации банков клеток.</p></sec><sec><title>Заключение</title><p>Заключение. В данной статье изложены основные принципы разработки, характеризации и трансфера банков клеток на основе бактериальной системы экспрессии.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Introduction</title><p>Introduction. Biological medicines produced using recombinant DNA technology play an important role in the international pharmaceutical market. Currently, bacterial and mammalian expression systems are widely used to produce recombinant proteins, monoclonal antibodies, and vaccines. To maintain the quality and stability of inoculation, cell banks are generated.</p></sec><sec><title>Text</title><p>Text. The general recommendations for cell bank development are outlined in the ICH Q5 guidelines. In the Russian Federation, similar requirements are specified in Decision No. 89 of the Council of the Eurasian Economic Commission "On approval of the Rules for assessments of biological medicines in the EAEU". Based on these requirements, one- or two-level cell banks should be established. Initially, a Master cell bank (MCB) is generated from a well-characterized single-cell bacterial colony according to high-quality standards. Subsequently, a Working cell bank (WCB) is created from one or several well-characterized MCB cryovials. As a result, the number of parameters required for WCB characterization can be reduced. The identity, purity, and stability of the cell bank should be determined. Requirements for cell bank characterization are detailed in official guidelines. An identity test is used to confirm the cell line or strain identification, which is a critical step to prevent cross-contamination and ensure the correct cell line in manufacturing processes. The absence of bacterial, fungal, or other types of contamination is demonstrated through purity testing. The sterility test is a key component, ensuring that a pure cell is used. The stability test demonstrates the genetic stability of the cells, including the preservation of genetic characteristics and the monitoring of harmful mutations during cultivation processes. Comprehensive data on the cell bank history, development, and characterization should be provided during its transfer.</p></sec><sec><title>Conclusion</title><p>Conclusion.
