BBP2023 – новый лекарственный кандидат группы сиднониминов, обладающий церебральной вазодилатирующей активностью и прокогнитивными свойствами. Фармакологические эффекты BBP2023 обусловлены наличием активных метаболитов. Для проведения фармакокинетических исследований на доклиническом этапе необходимо разработать биоаналитическую методику определения BBP2023 и его метаболитов в плазме крови кроликов.</p></sec><sec><title>Цель</title><p>Цель. Разработка и валидация хромато-масс-спектрометрической методики количественного определения нового производного сиднонимина и его метаболитов в плазме крови кроликов.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Количественное определение BBP2023 в плазме крови кроликов проводили методом ВЭЖХ-МС/МС. В процессе пробоподготовки биоматериала применяли способ осаждения белков метанолом. Хроматографическое разделение проводили смесью деионизированной воды и ацетонитрила в градиентном режиме на колонке ZORBAX Eclipse XDB-C18, 4,6 × 50 мм, 5 мкм, с помощью хроматографа Agilent 1260 Infinity II. Детектирование осуществляли в режиме MRM с использованием масс-спектрометра AB Sciex QTRAP 3200 MD. В качестве внутреннего стандарта применяли N-(этоксикарбонил)-3-(4-метилпентан-2-ил)сиднонимин.</p></sec><sec><title>Результаты и обсуждение</title><p>Результаты и обсуждение. Разработана методика пробоподготовки плазмы крови кроликов, подобраны оптимальные условия хроматографирования и детектирования исследуемого соединения и его метаболитов. Биоаналитическая методика валидирована по следующим показателям: селективности, матричному эффекту, степени извлечения, переносу пробы, линейности калибровочного диапазона, нижнему пределу количественного определения, внутри- и межсерийной точности и прецизионности, а также стабильности аналитов и внутреннего стандарта на этапах анализа.</p></sec><sec><title>Заключение</title><p>Заключение. Разработана и валидирована методика количественного определения нового производного сиднонимина BBP2023 и его метаболитов в плазме крови кроликов с применением ВЭЖХ-МС/МС. Подтвержденные аналитические диапазоны для BBP2023, BBP2023 А и BBP2023 С составили 5,0–500,0 нг/мл (5,0–1000,0 нг/мл для геранамина). Методика была использована при проведении аналитической части доклинического фармакокинетического исследования BBP2023 на кроликах.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Introduction</title><p>Introduction. BBP2023 is a new drug candidate of the sydnonymine group with cerebral vasodilating activity and procognitive properties. The pharmacological effects of BBP2023 are due to the presence of active metabolites. To conduct pharmacokinetic studies at the preclinical stage, it is necessary to develop a bioanalytical method for the determination of BBP2023 and its metabolites in rabbit blood plasma.</p></sec><sec><title>Aim</title><p>Aim. Development and validation of a chromatography-mass spectrometric method for the quantitative determination of a new derivative of sydnonymine and its metabolites in rabbit blood plasma.</p></sec><sec><title>Materials and methods</title><p>Materials and methods. Quantitative determination of BBP2023 in rabbit blood plasma was performed by HPLC-MS/MS. The biomaterial sample preparation was based on the method of protein precipitation with methanol. Chromatographic separation was performed with a mixture of deionized water and acetonitrile in a gradient mode on a ZORBAX Eclipse XDB-C18 analytical column (4.6 × 50 mm, 5 μm) using an Agilent 1260 Infinity II chromatograph. Detection was performed in the MRM mode using an AB Sciex QTRAP 3200 MD mass spectrometer. N-(ethoxycarbonyl)-3-(4-methylpentan-2-yl)sydnonymine was used as the internal standard.</p></sec><sec><title>Results and discussion</title><p>Results and discussion. The method for sample preparation of rabbit blood plasma has been developed, optimal conditions for chromatography and detection of the studied compound and its metabolites have been selected. The bioanalytical method has been validated according to the following parameters: selectivity, matrix effect, recovery rate, carry-over effect, linearity of the calibration range, lower limit of quantitative determination, intra- and inter-run accuracy and precision, as well as stability of analytes and the internal standard at the analysis stages.</p></sec><sec><title>Conclusion</title><p>Conclusion. A method for the quantitative determination of a new derivative of sydnonymine BBP2023 and its metabolites in rabbit blood plasma using HPLC-MS/MS has been developed and validated. The confirmed analytical ranges for BBP2023, BBP2023 A and BBP2023 C were 5.0–500.0 ng/ml (5.0–1000.0 ng/ml for geranamine).
