Определение возможности выработки нейтрализующих антител к инсулину, уменьшающих терапевтических эффект принимаемого препарата у пациентов, является важным этапом для понимания фармакологического профиля лекарственного препарата. Применение клеточных методов анализа на антитела позволяет проводить определение нейтрализующих антител к инсулину.</p></sec><sec><title>Цель</title><p>Цель. Адаптация и валидация методики оценки иммуногенности инсулина в плазме крови человека.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. В основе метода лежит использование клеточной линии iLiteTM Insulin Assay Ready Cells [<xref ref-type="bibr" rid="cit1">1</xref>], в геном которой встроен репортёрный ген люциферазы светлячка (Firefly) под контролем инсулинозависимого промотора. При увеличении концентрации инсулина экспрессия люциферазы светлячка (Firefly) увеличивается, что позволяет использовать данную клеточную линию для оценки количества нейтрализующих инсулин антител. Для нормализации по количеству клеток и учёта влияния матричного эффекта исследуемых образцов используется второй репортёрный ген люциферазы Renilla, который экспрессируется под контролем конститутивного промотора. Активность обеих люцифераз измеряли с помощью набора реагентов DualGlo Luciferase Assay System (Promega) [<xref ref-type="bibr" rid="cit2">2</xref>].</p></sec><sec><title>Результаты и обсуждения</title><p>Результаты и обсуждения. Были установлены оптимальные концентрация инсулина и степень разбавления плазмы/сыворотки для определения нейтрализующих антител к инсулину. Показана долгосрочная стабильность нейтрализующих антител к инсулину в плазме крови человека более 3-х месяцев. Описанная методика были применена при сравнительном исследовании безопасности и иммуногенности аналогов инсулина (Гларгин).</p></sec><sec><title>Заключение</title><p>Заключение. Разработана и валидирована методика определения антител к инсулину в K2EDTA плазме крови человека с помощью тестсистемы на основе клеточной линии iLiteTM Insulin Assay Ready Cells; в основе метода лежит оценка связывания альфа цепи инсулина с высокоаффинным гетеродимерным рецептором CD220.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Introduction</title><p>Introduction. IImmunogenicity identification of therapeutic proteins, such as human insulin analogues, is one of the most relevant and significant area in medicine and pharmaceuticals. Determination the possibility of producing neutralizing antibodies to insulin reducing the therapeutic effect of the drug, is an important step to understand the pharmacological profile of the drug. Applying of cell-based methods one allows to determinate neutralizing antibodies to insulin.</p></sec><sec><title>Aim</title><p>Aim. Development and validation methods for detection of neutralizing antibodies against insulin in human plasma.</p></sec><sec><title>Materials and methods</title><p>Materials and methods. The method is based on the use of the iLiteTM Insulin Assay Ready Cells [<xref ref-type="bibr" rid="cit1">1</xref>], in the genome of which the firefly luciferase reporter gene is introduced under the control of an insulin-dependent promoter. As the insulin concentration increases, the firefly luciferase expression (Firefly) increases, allowing one to use this cell line to estimate the number of neutralizing antibodies against insulin. For normalization by the number of cells and considering the matrix effect of studied samples, the second reporter gene luciferase Renilla is used, which is expressed under the control of a constitutive promoter. The activity of both luciferases was measured using the DualGlo Luciferase Assay System (Promega) assay [<xref ref-type="bibr" rid="cit2">2</xref>].</p></sec><sec><title>Results and discussion</title><p>Results and discussion. Optimal insulin concentration and plasma/serum dilution were determined to identify neutralizing antibodies to insulin. The long-term stability of neutralizing antibodies to insulin were shown in human plasma for more than 3 months. The developed method was applied in a comparative research of the safety and immunogenicity of insulin analogues (Glargine).
