Налтрексон – антагонист µ-опиоидных рецепторов является перспективным для лечения различных аутоиммунных и онкологических заболеваний при применении в дозах 1,5–5 мг/сут. На сегодняшний день лекарственные препараты, обеспечивающие такие дозировки налтрексона, отсутствуют. В связи с отсутствием на рынке препаратов, обеспечивающих такие дозировки налтрексона, нами был подобран состав назального спрея налтрексона гидрохлорида, содержащего водорастворимый термочувствительный полимер – полоксамер Kolliphor® Р 407. Для данного состава необходимо разработать методику количественного определения активного вещества с учетом сложности состава.</p></sec><sec><title>Цель</title><p>Цель. Разработка и валидация методики количественного определения налтрексона гидрохлорида в назальном спрее методом высокоэффективной жидкостной хроматографии (ВЭЖХ).</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. В качестве объекта исследования использовали назальный спрей налтрексона гидрохлорида. Методика количественного определения налтрексона в исследуемом образце была разработана на высокоэффективном жидкостном хроматографе «Dionex UltiMate 3000» (Thermo Scientific, США), оснащенном диодно-матричным детектором.</p></sec><sec><title>Результаты и обсуждение</title><p>Результаты и обсуждение. Изучена возможность применения изократических и градиентных режимов хроматографирования для количественного определения налтрексона гидрохлорида в назальном спрее. На основании этих результатов предложена новая методика определения с использованием градиентного режима, которая позволяет минимизировать влияние полимерного компонента в исследуемом образце на результаты анализа.</p></sec><sec><title>Заключение</title><p>Заключение. Разработана новая методика, позволяющая осуществлять идентификацию и определять количественное содержание налтрексона гидрохлорида в назальном спрее, содержащем в качестве загустителя водорастворимый термочувствительный полоксамер в высокой концентрации. Разработанная методика была валидирована по параметрам: правильность, прецизионность, специфичность, линейность.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Introduction</title><p>Introduction. Naltrexone, an antagonist of µ-opioid receptors, is promising for the treatment of various autoimmune and oncological diseases when used in doses of 1.5–5 mg/day. To date, there are no medications that provide such dosages of naltrexone.</p></sec><sec><title>Aim</title><p>Aim. Development and validation of a method for the quantitative determination of naltrexone hydrochloride in a nasal spray by high performance liquid chromatography (HPLC).</p></sec><sec><title>Materials and methods</title><p>Materials and methods. As an object of research, a naltrexone hydrochloride nasal spray was used. The quantitative determination of naltrexone in the test sample was developed using a Dionex UltiMate 3000 high-performance liquid chromatograph (Thermo Scientific, USA) equipped with a diode-matrix detector.</p></sec><sec><title>Results and discussion</title><p>Results and discussion. The possibility of using isocratic and gradient chromatographic modes for the quantitative determination of naltrexone hydrochloride in the nasal spray was studied. Based on these results, a new method of determination using the gradient mode is proposed, which allows minimizing the influence of the polymer component in the test sample on the analysis results.</p></sec><sec><title>Conclusion</title><p>Conclusion. A new technique of high-performance liquid chromatography (HPLC) is proposed that allows identification and quantification of naltrexone hydrochloride in a nasal spray containing a high concentration of water-soluble heat-sensitive poloxamer as a thickener.
