B-клеточная неоплазия плазмы клеток является причиной плазмоклеточной миеломы – второго по распространенности злокачественного заболевания системы крови. Помалидомид применяют в качестве терапии плазмоклеточной миеломы в случаях тяжелого течения заболевания и отсутствия эффективности прочих терапий. Помалидомид является структурным аналогом талидомида, он одобрен в России и США в качестве иммуномодулирующего препарата. В настоящее время данные по определению помалидомида в биологических жидкостях методом высокоэффективной жидкостной хроматографии с тандемным масс-спектрометрическим детектированием (ВЭЖХ-МС/МС) для исследований фармакокинетики практически отсутствуют, а имеющиеся отличаются по условиям детектирования. В данном исследовании приведена разработка и валидация методики определения помалидомида в плазме крови человека методом ВЭЖХ-МС/МС. В качестве пробоподготовки был выбран способ осаждения метанолом.</p></sec><sec><title>Цель</title><p>Цель. Целью исследования является разработка и валидация методики определения помалидомида в плазме крови человека методом ВЭЖХ-МС/МС для проведения фармакокинетических исследований.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Определение помалидомида в плазме крови проводили методом ВЭЖХ-МС/МС. В качестве пробоподготовки был использован способ осаждения белков метанолом.</p></sec><sec><title>Результаты и обсуждение</title><p>Результаты и обсуждение. Разработанная методика была валидирована по следующим валидационным параметрам: селективность, эффект матрицы, калибровочная кривая (линейность), точность, прецизионность, степень извлечения, нижний предел количественного определения, предел обнаружения, перенос пробы, стабильность.</p></sec><sec><title>Заключение</title><p>Заключение. Разработана и валидирована методика количественного определения помалидомида в плазме крови человека методом ВЭЖХ-МС/МС. Подтвержденный аналитический диапазон методики составил 1,00–500,00 нг/мл в плазме крови. Полученный аналитический диапазон позволяет применять разработанную методику для проведения аналитической части исследований фармакокинетики препаратов помалидомида.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Introduction</title><p>Introduction. B-cell malignancies of the plasma cell leads to the second most spread hematological malignancy disease, called multiple myeloma. Pomalidomide is used in case of previous multiple myeloma ineffective treatment. Pomalidomide is a thalidomide synthetic derived, approved as immunomodulatory drug by the Food and Drug Administration (FDA). Nowadays, detection of pomalidomide in blood plasma by high performance liquid chromatography tandem mass spectrometry (HPLC-MS/MS) is not spread. Moreover, the detection and the experimental setting accumulated data are varying greatly. This investigation provides development and validation of pomalidomide aiming to determine human blood plasma by HPLC-MS/MS method. The samples were processed by methanol protein precipitation.</p></sec><sec><title>Aim</title><p>Aim. The aim of this study is to develop a method for the pomalidomide in human plasma by HPLC-MS/MS for pharmacokinetic studies.</p></sec><sec><title>Materials and methods</title><p>Materials and methods. Determination of pomalidomide in plasma by HPLC-MS/MS. The samples were processed by methanol protein precipitation.</p></sec><sec><title>Results and discussion</title><p>Results and discussion. This method was validated by next parameters: selectivity, matrix effect, calibration curve, accuracy, precision, spike recovery, lower limit of quantification, detection limit, carry-over and stability.</p></sec><sec><title>Conclusion</title><p>Conclusion. The method of the determination of pomalidomide in human plasma was developed and validated by HPLC-MS/MS. The linearity in plasma sample was achieved in the concentration range of 1,00 – 500,00 ng/ml.
